Approaching the Glyphosate Herbicide: Structural, Electronic, and Vibrational Properties

O glifosato [n-(fosfonometil) glicina] é um herbicida não-seletivo cuja fórmula molecular é C3H8NO5P e é utilizado para controlar plantas daninhas (anuais ou perenes). Ele é absorvido pela folhagem e transportado por toda a planta, assim nenhuma parte da planta sobrevive. Atualmente, representa 60% do mercado mundial de herbicidas não seletivos, contabilizando um total de US$1,2 bilhão/ano em vendas, e é uma das melhores relações custo / benefício para o produtor rural, quando comparado aos outros métodos de controle de plantas daninhas. O glifosato atua inibindo a enzima EPSPS (5-enolpiruvato-chiquimato-3-fosfato sintase) que sintetiza os aminoácidos aromáticos fenilalanina, tirosina e triptofano, essenciais ao crescimento das plantas e não presente em mamíferos. Portanto, dependendo da dose, não é tóxico para esses seres. Além disso, o glyphosate possui propriedades favoráveis para o meio ambiente. Ele se liga firmemente ao solo, desta forma possui pouca mobilidade no solo, não atingindo águas subterrâneas. Também possui biodegradação microbiana em solo e água, alta solubilidade em água e é não volátil, portanto, não há contaminação atmosférica. Por outro lado, há evidências de efeitos deletérios no meio ambiente após o uso prolongado deste herbicida, principalmente devido à resistência ao glifosato adquirida por algumas espécies de ervas. Neste trabalho, utilizamos simulações computacionais para investigar as propriedades estrutural, eletrônica e vibracional do herbicida glifosato, pois é extremamente importante compreender como este herbicida funciona em nível molecular e como desencadeia os efeitos deletérios. Em particular, comparamos a teoria e os experimentos das propriedades vibracional e conformacional do glifosato com a variação do pH. Aqui, estudamos duas estruturas protonadas do glifosato: Gly-1 (pH=4.8) e Gly-2 (pH=8.0). Apresentamos os espectros infravermelho (IR) e o Raman que tornou possível a identificação dos confôrmeros para cada estado de protonação. Cálculos Ab initio foram feitos através da Teoria do Funcional da Densidade (DFT) com o funcional hibrido da densidade, o B3LYP, aplicando o conjunto de base 3-21G no vácuo e no meio aquoso, utilizando o modelo continuo de solvatação (PCM, Polarizable Continuum Model). Os resultados mostraram a eficiência da metodologia RPESS para uma busca racional de conformeros para o glifosato (Gly-1 e Gly-2), pois as conformações mais estáveis no modelo PCM são C1 e D1 e na fase gasosa são C2 e D3. O modelo PCM demonstrou ser importante para a estabilidade relativa da conformação C1. Foram obtidos os espectros teóricos para o Gly-1 e Gly-2. Os modos foram designados para todas as bandas entre 0 e 3600 cm-1.

The Glyphosate [n-(Phospometil) Glycine] is a non-selective herbicide, which the molecular form is C3H8NO5P and it has been utilized to control the weeds (yearly or perennial). It is absorbed by the foliage and it is transported to all the plant, in this way none part of the plant survives. Currently, represents 60% of the world market of non-selective herbicides, with a total of US$1.2 billion/year in the sales, and it is one of the best cost benefit relation for the rural producer, when compared to the other methods of weed control. The Glyphosate act restraining the EPSPS enzyme (5-enolpyruvate-chiquimate-3-phospate synthase) that synthesize the aromatic amino acids known as Phenylalanine, tyrosine, tryptophan, essential for the plant growing and absent in mammals. Therefore, depending on the dose, is not toxic for these beings. Beyond that, the Glyphosate has many favorable proprieties to the environment. It links firmly on the soil, and because of this, it has low mobility on it, without achieving the sub terrain water reserves. It has also microbial biodegradation in the soil, and also in the water, high water solubility and it is nonvolatile, that is why it has no environment contamination. On the other hand, there are evidences of the damaging effect on the environment, when this compound is utilized for a long time, mainly because of the glyphosate resistance acquired by some herbal species. In this project, we are using computational simulations to investigate the structural, electronic and vibrational proprieties of the glyphosate herbicide, because its extremely important to comprehend how this herbicide works at a molecular level and how it trigs the damaging effects. In particular, we compare the theory and the experiments of the vibrational and conformational proprieties of the glyphosate with the pH variation. Here, we studied two glyphosate protonated structures: Gly-1 (pH=4.8), Gly-2 (pH=8.0). We presented the infrared (IR) spectra and Raman that makes possible the identification of the conformers to the protonation state. Ab initio Calculations were made by the Density Functional Theory (DFT) with the hybrid functional of density, the B3LYP, applying the 3-21G base set in the vacuum and in the water, utilizing the Polarized Continuum Model (PCM). The results showed efficiency of a RPESS methodology for a rational search of conformers for the glyphosate (Gly-1 and Gly-2) is demonstrated, since the most stable C1 and D1 conformations in PCM model and C2 and D3 conformations in gas phase were found using this protocol. The PCM model demonstrated to be important for the relative stabilities of the conformation C1. The theoretical spectra of the glyphosate with the Gly-1 and Gly-2 were obtained. Modes were assigned to all bands between 0 and 3600 cm-1.
