SISTEMAS MICROFLUÍDICOS DE GOTAS PARA INCORPORAÇÃO DE DNA EM LIPOSSOMAS CATIÔNICOS PARA APLICAÇÃO EM TERAPIA GÊNICA

A terapia gênica visa à inserção de ácidos nucleicos em células-alvo, porém uma melhor eficiência de transfecção se dá com menor tamanho e polidispersidade dos complexos. Sistemas microfluídicos baseados em gotas são capazes de promover uma mistura rápida, gerando complexos com estas características. Neste contexto, utilizou-se no sistema microfluídico de gotas o óleo biocompatível FC-40 como fase contínua e a solução aquosa de lipossoma catiônico juntamente com pDNA, como fase dispersa. Incorporou-se pDNA nos lipossomas fixando o número de capilaridade (Ca), número adimensional que relaciona as forças viscosas e a tensão superficial entre as duas fases imiscíveis, em 0,003. Os lipossomas após incorporação do pDNA continuaram com o tamanho de aproximadamente 80 nm, a polidispersidade aumentou de 0,18 para 0,20 e potencial zeta diminuiu de 60 para 45 mV. Assim, este sistema nos permitiu obter complexos com propriedades desejáveis para transfecção.
